BAB I PENDAHULUAN
1.1 Latar Belakang
Prospek usaha tani jagung dan kedelai cukup cerah bila dikelola secara intensif dan komersial kepada agrobisnis. Permintaan pasar dalam negeri dan peluang ekspor komoditas jagung cenderung meningkat dari tahun ke tahun, baik untuk memenuhi kebutuhan pangan maupun nonpangan. Produksi dan kualitas jagung dapat ditingkatkan melalui penerapan teknologi mutakhir (modern) yang disesuaikan dengan agroekologi setempat. Rendahnya hasil rata-rata jagung nasional, anatara lain disebabkan belum meluasnya penanaman varietas - varietas vari etas unggul dan belum memperhatikan penggunaan benih
berkualitas
ditingkat petani,
dan disamping
itu,
pengelolaan
tanaman
dilingkungan budidaya tanaman jagung, misalnya teknik bercocok tanam, pemupukan, pengendalian hama dan penyakit, belum sesuai dengan paket teknologi maju yang berkembang di lapangan atau teknologi penelitian para pakar dibidangnya, hal inilah yang menjadi latar belakang usaha dalam upaya peningkatan produksi pertanian khususnya pertanian jagung di Indonesia. Pada dasarnya tanaman penyerbuk silang adalah heterozigot dan heterogenus. Satu individu dan individu lainnya genetis berbeda. Karena keragaman genetis yang umumnya cukup besar dibanding dengan tanaman penyerbuk sendiri dalam menentukan kriteria seleksi diutamakan pada sifat ekonomis yang terpenting dulu, tanpa dicampur aduk dengan sifat – sifat – sifat sifat lain yang kurang urgensinya. Pengertian yang bertalian dengan keseimbangan Hardy-Weinberg pengertian mengenai silang dalam, macam – macam gen dan sebagainya sangat membantu memahami sifat – sifat – sifat sifat tanaman penyerbuk silang dan metode – metode – metode metode seleksinya. yang dikembangkan secara aseksual. Beberapa metode pemuliaan tanaman yang diketahui yaitu introduksi, seleksi dan hibridisasi dilanjutkan seleksi. Metode pemuliaan tanaman ini punya manfaat yang sangat penting bagi perakitan varietas. Hibridisasi merupakan salah satu metode pemuliaan tanaman ta naman dimana bertujuan memperoleh kombinasi genetik yang diinginkan melalui persilangan dua atau at au lebih tetua yang berbeda genotipenya. Diharapkan setelah adanya hibridisasi dapat menghasilkan kombinasi baru genetika dari tanaman tetua yang diharapkan sifat unggulnya harapkan sifat unggulnya. Pemuliaan tanaman pada dasarnya adalah kegiatan memilih atau menyeleksi dari suatu populasi untuk mendapatkan genotipe tanaman yang memiliki sifat-sifat unggul yang selanjutnya akan dikembangkan dan diperbanyak sebagai benih atau bibit unggul. Namun demikian, kegiatan seleksi tersebut seringkali tidak dapat langsung diterapkan, dit erapkan, karena sifat-sifat keunggulan yang dimaksud tidak seluruhnya terdapat pada satu genotipe saja, melainkan terpisah pada genotipe yang lainnya. Misalnya, suatu genotipe mempunyai daya hasil yang tinggi tapi rentan terhadap penyakit, sedangkan genotipe lainnya memiliki sifat-sifat lainnya (sebaliknya). Jika seleksi diterapkan secara langsung maka kedua sifat unggul tersebut akan selalu terpisah pada genotipe yang berbeda. Oleh sebab itu untuk mendapatkan genotipe yang baru yang memiliki kedua sifat unggul tersebut perlu dilakukan penggabungan melalui rekombinasi gen. Persilangan merupakan salah satu cara untuk menghasilkan rekombinasi gen. Secara teknis, persilangan dilakukan dengan cara memindahkan tepung sari ke kepala putik pada tanaman yang diinginkan sebagai tetua, tet ua, baik pada tanaman yang menyerbuk sendiri (self polination crop) maupun pada tanaman yang menmyerbuk silang (cross polination crop).

Jagung termasuk tanaman berumah satu, dimana bunga jantan dan bunga betina terpisah tetapi terdapat pada satu tanaman. Malai merupakan rangkaian bunga jantan yang terletak pada ujung batang. Malai bercabang-cabang, pada tiap cabang terdapat deretan spikelet yang masing-masing terdiri dari sepasang bunga jantan. Tiap bunga jantan mempunyai 3 kepala sari (anther), masing-masing menghasilkan tepung sari sampai 12 juta butir. Tepung sari sangat lembut dan ringan sehingga mudah tersebar oleh angin. Penyebaran tepung sari biasanya sejauh 6-15 m tetapi dapat mencapai 800 m. Tongkol muda yang biasanya tumbuh pada ruas batang ke-6 sampai ke-8 dari pangkal, merupakan rangkaian bunga betina. Pada janggung muda tersusun pasangan pasangan bunga betina, tetapi teta pi hanya bunga disebelah atas dari masing-masing pasangan yang terus hidup. Rambut jagung adalah putik yang ujungnya bercabang dan penuh dengan bulu halus. Bulu halus jumlahnya semakin berkurang mendekati pangkal. Untuk pembuahan, tepung sari tumbuh kedalam putik melalui bulu tersebut. Satu tongkol hanya memerlukan paling banyak 800 - 1000 tepung sari. Jagung adalah tanaman menyerbuk secara silang. Penyerbukan terjadi terutama oleh bantuan angin. Dilapang terjadi 97 % atau lebih penyerbukan silang antara tanaman satu dengan yang lain. Daun jagung tumbuh melekat pada buku-buku batang, struktur daun terdiri atas tiga bagian yaitu kelopak daun, lidah daun(ligula) dan helaian mdaun. Mbagian permukaan daun berbulu, dan terdiri te rdiri dari sel-sel s el-sel bulifor. bulif or. Jumlah helaian daun bervariasi antar 8-48 helaian. Bunga jantan matang terlebih dahulu 1-2 hari dari pada bunga betian. Buah jagung terdiri atas tongkol, biji dan daun pembungkus. B iji jagung mempunyai bentuk , warna dan kandungan endosperm yang bervariasi tergantung jenisnya. Biji jagung terdiri atas tiga bagian utama yaitu kulit biji (seedcoat), endosperm, dan embrio (Hasym,2007). Faktor-faktor yang paling penting dalam penanaman jagung antara lain sinar matahari, air, hujan dan angin. Air yang memadai di daerah areal sekitar pertanian yang cukup akan membantu biji, bunga, dan buah dalam proses pertumbuhan dan disertai hujan yang relative optiamal. Keberadaan angin juga sangat penting didalam membantu penyerbukan. Temperature untuk jagung berkisar antara 23-27 0C (Allard, 1992). Tanaman jagung dapat hidup dengan pH berkisar antara 5,5- 7,0. keasaman tanah sangat erat didalam pembentukan biji, buah serta pertumbuhan tanaman. Jenis tanah yang baik untuk jagung berupa tanah yang gembur dan remah-reamah. Tekstur tanah seperti lempung berdebu atau berpasir. Disisni juga diperlukan aerase yang baik untuk meningkatkan peredaran udar didalam tanah(Rukmana,1997). Untuk tanaman jagung kemirigan tanah optimum adalah 5-8 %. Hal ini dikarenakan kemungkinan terjadinya erosi tanah sangat rendah. Pada daerah dengan tingkat kemiringan lebih dari 8 % kurang sesuai untuk penanaman jagung (Warisno, 1998). langsung melalui persaingan antar tanaman dalam memanfaatkan faktor tumbuh. Jarak tanaman yang tepat tajuk tanaman akan segera menutup yang secara tidak langsung akan menghambat pertumbuhan gulma sehingga pertumbuhan tanaman akan lebih baik yang akhirnya dapat memberikan hasil yang tinggi (Bunting, 1973)
2.2 Hibridisasi
Hibridisasi ialah perkawinan antara berbagai spesies, suku, ras atau varietas tumbuhan yang bertujuan memperoleh organisme yang diinginkan. Tujuan hibridisasi untuk menambah keragaman genetik melalui proses pengkombinasian genetik dari tetua yang berbeda genotipnya. Dari tujuan tersebut dapat diketahui bahwa hibridisasi memiliki peranan penting dalam pemuliaan tanaman, terutama dalam memperluas keragaman genetic (Purnamasari, 2012) Penyerbukan adalah jatuhnya serbuk sari ke kepala putik. Sedangkan pembuahan adalah bergabungnya gamet jantan dan gamet betina. Kriteria klasifikasi yang dipergunakan hanya berdasarkan tingkat penyerbkan sendiri dan penyerbukan silang. Polonasi sendiri sudah barang tentu hanya merupakan salah satu system perbanyakan tanaman dan hanya sebagai salah satu jalan dimana populasi dapat dikawinkan. Didalam group penyerbukan silang jumlah persilangan dari luar adalah sangat penting karena ia memepengaruhi dalam kontaminasi stok pemuliaan. Ada perbedaan yang besar antara jumlah persilangan dengan luar didalam species dari suatu kelompok. Jumlah persilangan dari varietas yang diberikan juga dipengaruhi oleh keadaan lingkungan yang berubah (Allard, 1998). Terjadinya penyerbukan silang disebabkan oleh : a. Metode pemuliaan tanaman menyerbuk silang sedikit berbeda dengan tanaman menyerbuk sendiri karena pada tanaman menyerbuk silang, dalam populasi alami terdapat individu-individu yang secara genetik heterozigot untuk kebanyakan lokus. Secara genotipe juga berbeda dari satu individu ke individu lainnya, sehingga keragaman genetik dalam populasi sangat besar. Fenomena lain yang dimanfaatkan dalam tanaman menyerbuk silang adalah ketegaran hibrida atau heterosis. Heterosis didefinisikan sebagai meningkatnya ketegaran (vigor) dan besaran F1 melebihi kedua tetuanya. Sebaliknya bila diserbuk sendiri akan terjadi tekanan inbreeding. Beberapa metode yang populer pada tanaman menyerbuk silang misalnya pembentukan varietas hibrida, seleksi massa, seleksi daur ulang, dan dilanjutkan dengan pembentukan varietas bersari bebas atau varietas sintetik. sint etik. 2.3 Teknik persilangan persilangan pada kedelai kedelai
Kedelai merupakan tanaman semusim yang sering dibudidayakan petani selain tanaman padi. Seperti tanaman pada umumnya kedelai akan berbunga ketika memasuki masa generatifnya (kira-kira 40 HST). Putik bunga sangat kecil berukuran sekitar 3 mm dan berbentuk menyerupai calon polong. Setelah pembuahan terjadi putik tidak akan gugur dan berkembang menjadi polong kedelai. Benangsari bunga kedelai juga berukuran sangat kecil. Tangkai benangsari menyatu dan membentuk suatu selaput tipis yang menutupi/mengelilingi putik. Selaput ini mendukung benangsari diatasnya.

2.4 Tahap Persilangan Kedelai
Alat yang harus dipersiapkan untuk menyilangkan bunga adalah pinset, benang plastik penutup putik dan label. Secara umum proses persilangan bunga kedelai sama dengan teknik persilangan biasa. 1. Kastrasi Kastras i pada bunga kedelai cukup sulit dilakukan karena bunga berukuran kecil (sekitar 5-7mm ketika mekar sempurna) juga karena tangkai benangsari yang saling melekat dan membentuk membentuk seludang (selaput) menutupi putik. Alat yang digunakan dalam hibridisasi kedelai adalah pinset, benang berwarna, label dan pensil. Sementara untuk hibridisasi jagung antara lain kantong penutup tasel ukuran 5.75 × 4.25 × 14 (Lawson bag No.402), kantong penutup tongkol ukuran 2 × 1 × 7 (Lawson bag No.217), spidol permanen, staples, pisau, pensil dan label.

Pada setiap buku sebaiknya hanya 2 bunga saja yang disilangkan, sisanya dibuang. Kuncup bunga yang akan dikastari dipegang pangkalnya dengan ibu jari dan telunjuk tangan kiri. Penyerbukan dilakukan antara tanaman jagung yang berbeda varietas (crossing) atau antara bunga jantan dan betina pada varietas yang sama ( selfing ( selfing ). ). Pilih bunga betina (tongkol) yang akan diserbuki sebelum rambut pada ujung tongkol keluar, tongkol dibungkus dibungkus dengan kantong kantong kertas yang sudah sudah disiapkan. Penutupan bunga bunga betina dilakukan setiap hari. Pilih tanaman yang akan dipakai sebagai pejantan (sumber serbuk sari) dengan tanda-tanda bunga jantan sudah mekar, kemudian bungkus bunga jantan tersebut sampai rapat dengan kantong kertas, jangan sampai serbuk sari jatuh beterbangan. Setelah satu atau dua hari bunga jantan tersebut telah siap untuk disilangkan. Untuk memastikan diperolehnya tepung sari yang cukup, maka tepuklah bunga jantan yang terbungkus tersebut. Apabila bunga betina yang dipilih telah siap diserbuki, yaitu pada tongkol yang telah keluar rambut diujungnya, maka persilangan telah siap dilaksanakan. Persilangan dilakukan dengan cara memindahkan bunga jantan (serbuk sari) ke bunga betina (putik) dengan meletakkan serbuk sari pada rambut tongkol. Tongkol yang telah diserbuki ditutup kembali dengan kantong kertas yang digunakan untuk menutup tasel (bunga jantan). Tulis dan gantungkan label persilangan pada tongkol tersebut.
