Evolution and the functional significance of bacteriophages in the evolution of bacteria have intensively been studied over many years, although only a small number have been investigated in detail. Among 11 lambdoid prophages on the EHEC O157:H7 Sakai genome, Sp5 and Sp15 encode Stx2 and Stx1, respectively. Although the early region genes of Sp5, mostly involved in DNA replication and gene regulations, have high similarity with those of lambdoid phages, but many of late region genes that are presumably associated with virion formation show no sequence similarity to known phage genes, and thus their functions are largely unknown. Therefore, to understand these unknown genes’ roles in Sp5 propagation, we first constructed an O157:H7 Sakai derivative, in which the stx2 gene on Sp5 was replaced by the
-cassette, cat -cassette,
and lysogenized into E. coli strain K-12. Subsequently, a series of
Sp5 mutants were constructed under the K-12 background, each of which contains an in-frame deletion of the respective ORF in order to analyze their function(s). We also developed a modified double agar overlay method using Mitomycin C and CaCl 2 (or MgCl2) that allows us to easily detect and accurately enumerate plaques of Sp5. The results of systematic analysis of 38 mutants for their DNA-packaging, infection and plaque formation abilities indicated that several of these genes are required for or involved in the process of Sp5 propagation. Complementation analyses with wild type genes confirmed their essentiality in virion production and/or their infectivity, although complementation was unsuccessful in some of the mutants. We are now constructing additional mutants to cover all unknown genes of Sp5, including those in the early region, for better understanding the biology of Stx2-transducing phage of EHEC O157:H7. My research interests and activities over the past six years have mainly centered on the molecular study of the evolution of pathogenic intestinal bacteria and better understand the functional significances of bacteriophages in the evolution of bacteria. In addition to these research areas, I was also actively involved in the finding of naturally occurring molecule that might reduce the severity and the pathogenicity of gut pathogen i.e., Enterohemorrahgic Escherichia Escherichia coli (EHEC). Since I have a lot of experimental data supporting my goal therefore, with this study I have a plan to develop an antidote that can be use as the treatment of EHEC infection. Beside this I have also the keen interest in the study and elucidation of the molecular mechanism of multidrug resistant bacteria in Bangladesh. Currently, I am working as an assistant Professor in the Department of Biochemistry and Molecular Biology, University of Dhaka, Bangladesh with the key responsibilities of teaching and research. Since YSP-program is aimed at bringing young scientists from FAOBMB regions to come together for discussion and exchange of knowledge and ideas in relation to their research in the field of Biochemistry and Molecular Biology, therefore I strongly believe that my participation will help me to enrich my knowledge and skill in the field of molecular biology and I could share this with the student at the university level. Beside this I will be able to gain some ideas in current research trend and the residing problems in the Asian and Oceanian countries by exchanging the views with the young participants. My experiences in molecular biology will enable me to collaborate with clinicians, local and international scientists sci entists to conduct transdisciplinary studies in basic science and might help in our national developments. My participation will also help me immensely and boost up to run educational arts activities and events in my own county. Moreover, along with my teaching I aspire to become an independent researcher and have my own lab therefore, this YSP- program participation will help me a lot to achieve my goals.
I will show my research data and the expertise that I have in the field of molecular biology will be shared with the young scientists from all over the Asian and Oceanian region. I strongly believe that this scientific sharing will help many of the young participants that in turn will help for their national developments.
