Safety Reminder WARNING 1. This centrifuge is not explosion-proof. Never use explosive or flammable samples. 2. Do not install the centrifuge in or near places where flammable gases are generated or chemicals are stored. 3. Take all necessary safety measures before using samples that are toxic or radioactive, or blood samples that are pathogenic or infectious. You use such samples at your own responsibility. 1) If the centrifuge, rotor, or an accessory is contaminated by samples that are toxic or radioactive, or blood samples that are pathogenic or infectious, be sure to decontaminate the item according to good laboratory procedures and methods. 2) If there is a possibility that the centrifuge, rotor, or an accessory is contaminated by samples that might impair human health (for example, samples that are toxic or radioactive, or blood samples that are pathogenic or infectious), it is your responsibility to sterilize or decontaminate the centrifuge, rotor, or the accessory properly before requesting repairs from a Hitachi Koki authorized sales/service representative. 3) It is your responsibility to sterilize and/or decontaminate the centrifuge, rotor, or parts properly before returning them to a Hitachi Koki authorized sales/service representative. 4. Because the liquid samples, the saline, etc. are used in this centrifuge, your centrifuge must be grounded properly. 5. To avoid electrical shocks, do not handle the power cord or turn on or off the POWER switch with wet hands. 6. For safety purposes, do not go within 30 cm of this centrifuge while it is in operation. 7. While the rotor is rotating, never forcedly unlock the door. 8. Repairs, disassembly, and other modifications to the centrifuge are strictly prohibited unless performed by a Hitachi Koki authorized sales/service representative.

Safety Reminder CAUTION 1. Do not move or relocate this centrifuge while the rotor is rotating. 2. Do not pour any solution such as water, detergent, or disinfectant directly into the rotor chamber. If you do so, the bearings of the drive unit might corrode or deteriorate. 3. Connect the saline tank to the pump inlet connector of the centrifuge properly with the hose bands (standard accessories). Otherwise the liquid might leak from the tube and the liquid might seep into the inside of the centrifuge. If the liquid might have seeped into the inside of the centrifuge, contact a Hitachi Koki authorized sales/service representative for cleaning and drying the centrifuge. 4. Before running this centrifuge, remove any dropped objects and tube fragments from inside the rotor chamber. 5. Always check for corrosion and damage on the rotor before using it. Do not use the rotor if you find such a problem. 6. Be sure to use the specified test tubes with a wall thickness of 1 mm or more. When using thin test tubes such as disposable test tubes, do not use them repeatedly. If any of scratches, crack, inside warp, etc. is found on the test tubes, do not use them because they cannot bear a centrifugal force. 7. Be sure to load test tubes to all the holders. When test tubes are not loaded unavoidably to all the holders, load test tubes and empty tubes to all the holders. 8. Use one or two drops (about 50μl) of 3-5% erythrocyte suspension as the sample volume at blood cell washing. When using precipitated erythrocyte layer, use one or two drops (about 50μl or less) per a test tube. Use the 80% or less of the test tube capacity as the sample volume for centrifugation. 9. If you observe some abnormality in this product, stop using it immediately and contact a Hitachi Koki authorized sales/service representative. Notify the service representative of the alarm code if displayed. 10. If the centrifuge will not be used for a long time, remove the power cord from the receptacle. 11. For connection to a different outlet, the power cord might be needed to be replaced. Follow local electrical codes. 12. Depending on the magnitude, an earthquake might damage the centrifuge. If you observe some abnormality, contact a Hitachi Koki authorized sales/service representative.
Note: During operation, it is not abnormality that the control panel and the surface of this centrifuge are warmed by themselves.

1. Operation Principle The MC450 automatically carries out a series of blood cell washing processes of erythrocyte suspension for antiglobulin test (direct Coomb’s test or indirect Coomb’s test). The operational sequence is shown below. ) m p r ( d e e p s r o t o R

WASH
A fixed amount of saline is pumped into the distributor when the rotor speed reaches 1200 rpm. By centrifugal force, the saline is jetted from the distributor into test tubes.

The rotor is rotated at a low speed with the rotor holder kept at an angle to slightly open its top end by magnetic force.

AGITATE
The rotor repeats rotation and stop at short, quick intervals to disentangle the remained blood cells.
⑤
CENTRIFUGE
The rotor is rotated about 5 seconds to collect the blood cells adhered to the wall surfaces of the test tubes to the bottom.
It is to ensure the reaction
with the Coombs reagent.

Function Shows the current process of blood cell washing. Also indicates the process setting in the case of manual mode. While the centrifuge is in operation, the LED for the process concerned blinks. Indicates the remaining time of centrifugation (in 3 digits). In case of an abnormality, it shows the error code concerned. It also shows the rotor speed (×10 rpm) when the PUSH SPEED switch on the condition-setting panel is pressed. To be pressed for automatic washing cycle in succession. Its LED lights when the automatic mode is set. To be pressed when executing a single process (WASH, CENTRIFUGE, DECANT or AGITATE) by manual control. Its LED lights when the manual mode is set. Function To be used for pump deaeration at the time of initial operation. The pump for saline supply operates while this switch is kept depressed. (This switch is ineffective while the centrifuge is in operation.) Sets the operation program. 7. Preparation for Operation 7.1 Preparation (1) Prepare the test tubes (10 or 12 mm in diameter and 75 mm in length). When using the test tubes 10 mm in diameter, set the attached D10 a dapter to the rotor holder. CAUTION: ● Be sure to use the specified test tube with a wall thickness of 1 mm or more. When using thin test tubes such as disposable test tubes, do not use them repeatedly. If any of scratch, crack, inside warp, etc. is found on the test tubes, do not use them because they cannot bear a centrifugal force. It is recommended to use the tempered hard glass test tubes made by NICHIDEN-RIKA GLASS CO., LTD. or the Pyrex test tubes made by ASAHI TECHNO GLASS CORPORATION for this centrifuge. (2) Prepare the saline tank (standard accessory) and a waste liquid tank. Connect the signal cord of the liquid level sensor of the saline tank to the connector at the rear of the centrifuge. (3) Perform piping as shown in the figure. ・Do not set the saline tank at a place higher than the centrifuge or behind the centrifuge. ・Set the waste liquid tank at a place lower than the centrifuge. ・Connect the drain pipe to the drain connector. ・Place the tips of the drain pipe in the waste liquid tank or a sink positioning them higher than the
liquid level. The gray connector at the rear center of the centrifuge is used for draining off waste liquid from the chamber. After blood cell washing, waste liquid keeps draining off through the connector. The auxiliary drain tube under the connector is an auxiliary tube to drain waste liquid flowed from the chamber into the vicinity of the driving motor. Although waste liquid may not always drain off through it, prepare a tank for it. CAUTION: ● Make sure that the tips of the drain pipes are above the waste liquid level in the tank. Otherwise, the chamber cannot be drained completely and waste liquid stagnates in the chamber due to poor drainage. Poor drainage may also be caused if the pipe is bent or flattened or if waste liquid stagnates in the middle of the pipe as shown by the dotted lines in the following figure. Set the pipes properly so that waste liquid drains off smoothly. Be careful not to set the drain pipes at a place higher than the drain outlet of the centrifuge. ●Connect the saline tank to the pump inlet connector of the centrifuge properly with the hose bands (standard accessories). Otherwise the liquid might leak from the tube and the liquid might seep into the inside of the centrifuge. If the liquid might have seeped into the inside of the centrifuge, contact a Hitachi Koki authorized sales/service representative for cleaning and drying the centrifuge. ・Connect the saline tank to the pump inlet connector of the centrifuge by tubing. 7.2
Injection Volume Adjustment and Pump Deaeration
(1) Adjustment of injection volume of saline The MC450 is factory-adjusted to the injection volume of saline for the 12-mm diameter test tubes. When using the 12-mm diameter test tubes with this centrifuge, there is no need to adjust the injection volume. When using the 10-mm diameter test tubes, adjust the flow rate of the pump as follows:
① Remove the protective cover of the pump at the rear of the centrifuge (it is secured with two screws). ② Loosen the two locking screws of the pump. ③ Turn the adjusting screw counterclockwise to set the indicator to “60%” position. ④ Retighten the two locking screws.

(2) Pump deaeration Fill the saline tank with saline. Hold a beaker or other container to the nozzle. Press the SALINE PRIME switch on the condition-setting panel to discharge saline from the nozzle until no bubbles come out of the nozzle.

7.3
Setting of Bowl, Rotor and Distributor
(1) Bowl setting
① Apply silicone grease (standard accessory, general vacuum grease is also usable) to the inside of the bowl (see the arrow mark in the figure) and the loading portion of the O-ring of the decantation coil lightly by hand. Perform grease application once or twice a month.

②Put both hands on the inside of the bowl as shown in the figure and push down the bowl horizontally Bowl applying a force with the thumbs and the other fingers alternately.

③When the bowl is pushed about one-third or a half of the bowl height, put both hands as shown in the figure and push down the bowl furthermore turning the bowl until the bottom of the decantation coil becomes flush with the bottom of the bowl. ④Check that the bowl can be turned smoothly by hand.

(2) Rotor setting Properly place the rotor on the drive shaft. Turn the rotor about a quarter so that the coupling pins are inserted into the rotor shaft holes.

Coagulation of blood cells is influenced by the centrifugal speed (centrifugal force) and the centrifugation time. If the centrifugal force and the time are insufficient, coagulation may also be insufficient causing false negative results. To the contrary, excessive centrifugation may harden the blood cells firmly causing false coagulation or difficult resuspension. Thus the weak reaction may disappear. In addition, centrifugal conditions are greatly influenced by the amount of blood cells, specific gravity of blood cells, etc. Note that the above setting is just as a guide. Determine the optimum conditions according to the following evaluation. ① The supernatant is transparent and no blood cell is suspended after centrifugation. ② The blood cells that are precipitated at the bottom of the test tubes show clear outlines after centrifugation. ③ The blood cells are easily removed from the bottom and disentangled by a light agitation after decantation. ④ Check the reaction using reagents whether negative or positive. 7.5 Preparation of the sample CAUTION: ●When the sample or the liquid is spilled, it might seep into the inside of the centrifuge. 8. Operation CAUTION: ●How to close the door Close the door by pushing down at the forward center of the door strongly until a short beep is heard. If the door is closed insufficiently, an alarm (E1: DOOR OPEN) is indicated and the centrifuge does not start operation. If this alarm is indicated, properly close the door and press the START switch again. ●How to open the door Open the door gently after blood cell washing. Otherwise the remaining waste liquid from the inside of the drain cover might splash.

Note that Hitachi Koki cannot repair the centrifuge, rotor or the accessory unless sterilization or decontamination is completed. ●It is your responsibility to sterilize and/or decontaminate the centrifuge, rotor, or parts properly before returning them to a Hitachi Koki authorized sales/service representative. In such cases, copy the decontamination sheet at the end of this manual and fill out the copied sheet, then attach it to the item to be returned. Hitachi Koki may ask you about the treatment for t he centrifuge, rotor or the part if the decontamination is checked and judged as insufficient by Hitachi Koki. (1) Storage period of service parts Service parts are kept in stock for seven years after the discontinuation of production. The term “service parts” means the parts that are necessary to ensure the correct functioning of the centrifuge.

9.1 Maintenance of Pump, Tank and Tubing ・
Wash the inside of the tank and the tubing once a week. If saline is contaminated by bacteria, it may cause hemolysis or poor result. If the centrifuge is not used for a long period, drain out saline from the tank, tubing and pump. Check the volume of saline inside the tank before operation. Check that the tube is connected properly. When the tube is not connected properly, connect the tube securely to the saline tank and the pump inlet connector with the hose bands (standard accessories). When th e tube is lax, replace it to the new one. Replace the drain pipe and the tube with new ones every 1 to 3 years depending on the degree of stains.

<Washing> ① Fill up the saline tank with washing solution (2% Sodium Hypochlorite Solution). Fill up the fluid passage with washing solution according to the same procedure as pump ② deaeration (see “7.2 Injection Volume Adjustment and Pump Deaeration”) and leave it for 12 to 24 hours. ③ After 12 to 24 hours, drain out washing solution from the saline tank and fill up the tank with distilled water. ④ Flow distilled water 2 to 3 liters from the saline tank according to the same procedure as pump deaeration. ⑤ Exchange distilled water in the saline tank for saline. ⑥ Flow saline 2 to 3 liters from the saline tank in the same manner as ④. The pump is a consumable part. Liquid may leak from the bellows of the pump due to deterioration. Although deterioration depends on the installation environment such as ultraviolet rays and temperature, it is recommended to replace t he bellows of the pump with new one (part number S403754) once every three years. It is also recommended to use saline that includes antiseptics.

9.2 Maintenance of Rotor, Distributor and Bowl After operation, remove the rotor, distributor and bowl properly, then wash and dry them well. See the following table for sterilization. Part

9.3 Maintenance of Chamber, Inner Chamber and Drain Cover Remove the inner chamber and the drain cover from the centrifuge properly, then wash and dry them well. A 2% sodium hypochlorite solution is used for sterilization. When washing the inside of the chamber, be careful not to pour washing solution on the decantation coil directly. Wipe the decantation coil with a cloth or paper moistened with washing solution.

9.3.1 Removing and Mounting the Drain Cover ①
Slide the knob of the lock fastener at both sides in the arrow direction.

9.3.2 Removing and Mounting the Inner Chamber ①

CAUTION: ●Do not store washing solution in the chamber. Do not hose on the decantation coil. Otherwise, liquid may enter the inside of the centrifuge causing a failure.
・If a test tube is broken, be sure to remove the fragments inside the chamber and the drain pipe of the drain cover thoroughly with a brush.

10.Troubleshooting 10.1 Error code If any abnormality occurs during operation, corresponding error code blinks on the TIME indicator and the alarm buzzer sounds, then the centrifuge stops. Take the action described in Table 10-1 or contact a Hitachi Koki authorized sales/service representative for repair.

In the manual mode, CENTRIFUGE and DECANT processes can be executed even when this error code is shown. The liquid level sensor may be faulty if the alarm buzzer sounds even though there is enough saline. Contact a Hitachi Koki authorized sales/service representative for repair. The error code can be temporarily cleared by pressing MAUAL and STOP switches simultaneously for three seconds or more (operation in the automatic mode is possible). It returns to the original state by turning on and off the power switch. If a power failure occurs while the centrifuge is running, wait a minute as the centrifuge stops rotation after the power failure. Press any of the switches on the control panel after a lapse of one minute or more from when the error code is indicated. If the error persists, contact a Hitachi Koki authorized sales/service representative for repair. This error may be caused by voltage drop when the rotor accelerating if your centrifuge is connected to an outlet to which multiple appliances are connected. Turn off the centrifuge power and connect the centrifuge to a proper outlet that can supply stable voltage to the centrifuge. The frequency of the power supply is abnormal. Turn off the power switch and on again. If the error persists, contact a Hitachi Koki authorized sales/service representative for repair. 11.Installation 11.1 Location (1)Install the centrifuge in a room where the ambient temperature is 5 to 30℃. (2)Place the centrifuge on a sturdy desk or a wagon with wheel locks or a strongly structured
workbench in a horizontal position. (3) Avoid installing the centrifuge in a dusty place. Otherwise, dust may get into the sample. (4)Do not position this centrifuge in direct sunlight. Otherwise, the temperature rises excessively and it may cause a failure. (5)Use the centrifuge in a well-ventilated room. Keep spaces at least 10 cm from the walls of the room. ( 6 ) This centrifuge is not explosion-proof. Do not use the centrifuge in or near places where flammable gases are generated or chemicals are stored.

11.4 Connection of the Power Cord WARNING: ●To avoid electrical shocks, do not touch the power cord with wet hands. ●Do not hold the cord when disconnecting the power cord from the outlet. Hold the plug when disconnecting it. ● This centrifuge must be grounded properly. The power cord plug has a three-prong plug. One of the three prongs is provided for grounding. This centrifuge can be grounded just by putting the three- prong plug into a three-prong outlet. Do not connect the three-prong plug to a two-prong outlet using an adapter.
CAUTION: ●For connection to a different outlet, the power cord may be needed to be replaced. Follow local electrical codes.

12. Opening the Door during Power Failure WARNING: ●Opening the door while the rotor is rotating is very dangerous. Never unlock the door while the rotor is rotating. ●In the event where the door is opened while the rotor is still rotating, close the door immediately. ●Do not run the centrifuge with the hex. wrench inserted in the hole of the centrifuge.
The door cannot be opened and closed except when the centrifuge is powered on (POWER switch is on) and the rotor is stopped. If the door cannot be opened due to a power failure, open the door according to the following procedure. (1) Check that the rotor is not rotating. ■Listen carefully to make sure that no rotating sound is heard. ■It takes about 30 seconds until the rotor rotating at 3,000 rpm (maximum speed)
stops completely.
